Phosphatidylinositol-phospholipase C4 suppresses the hypersensitive response of <i>Nicotiana benthamiana</i>

چکیده

Phospholipid signaling plays an important role in plant immune responses. Here, we isolated two phospholipase C4 (PLC4) orthologs the Nicotiana benthamiana genome, designated as N. PLC4-1 and PLC4-2 (NbPLC4-1 NbPLC4-2). We created NbPLC4-1- NbPLC4-2- silenced plants. Induction of hypersensitive response (HR), including HR cell death bacterial population reduction, was accelerated both NbPLC4-2-silenced plants challenged with benthamiana-incompatible Ralstonia solanacearum 8107. The also showed enhanced expression Nbhin1, a marker gene. Expressions genes for salicylic acid (SA) jasmonic (JA) were drastically increased by R. inoculation. In addition, NbPLC4-1 NbPLC4-2 silencing triggered reactive oxygen species (ROS) hyper-production. These results suggest that NbPLC4s are closely associated JA, SA, ROS act negative regulators benthamiana.

Phosphatidylinositol-specific phospholipase C of Bacillus anthracis down-modulates the immune response.

Phosphatidylinositol-specific phospholipases (PI-PLCs) are virulence factors produced by many pathogenic bacteria, including Bacillus anthracis and Listeria monocytogenes. Bacillus PI-PLC differs from Listeria PI-PLC in that it has strong activity for cleaving GPI-anchored proteins. Treatment of murine DCs with Bacillus, but not Listeria, PI-PLC inhibited dendritic cell (DC) activation by TLR l...

Non-specific phospholipase C4 mediates response to aluminum toxicity in Arabidopsis thaliana

Aluminum ions (Al) have been recognized as a major toxic factor for crop production in acidic soils. The first indication of the Al toxicity in plants is the cessation of root growth, but the mechanism of root growth inhibition is largely unknown. Here we examined the impact of Al on the expression, activity, and function of the non-specific phospholipase C4 (NPC4), a plasma membrane-bound isof...

Formation of oxidized phosphatidylinositol and 12-oxo-phytodienoic acid containing acylated phosphatidylglycerol during the hypersensitive response in Arabidopsis.

Plant membranes are composed of a wide array of polar lipids. The functionality of these extends far beyond a pure structural role. Membrane lipids function as enzyme co-factors, establish organelle identity and as substrates for enzymes such as lipases and lipoxygenases. Enzymatic degradation or oxidation (enzymatic or non-enzymatic) of membrane lipids leads to the formation of a diverse group...

AtCYS1, a cystatin from Arabidopsis thaliana, suppresses hypersensitive cell death.

In plants, cysteine protease inhibitors are involved in the regulation of protein turnover and play an important role in resistance against insects and pathogens. AtCYS1 from Arabidopsis thaliana encodes a protein of 102 amino acids that contains the conserved motif of cysteine protease inhibitors belonging to the cystatin superfamily (Gln-Val-Val-Ala-Gly). Recombinant A. thaliana cystatin-1 (A...

Phospholipase C Regulation of Phosphatidylinositol 3,4,5-trisphosphate-mediated Chemotaxis□V

Generation of a phosphatidylinositol 3,4,5-trisphosphate [PI(3,4,5)P3] gradient within the plasma membrane is important for cell polarization and chemotaxis in many eukaryotic cells. The gradient is produced by the combined activity of phosphatidylinositol 3-kinase (PI3K) to increase PI(3,4,5)P3 on the membrane nearest the polarizing signal and PI(3,4,5)P3 dephosphorylation by phosphatase and t...
